Abstract In the present investigation, the objectives were to optimize the micropropagation of black baccarat cultivar. The nodal segments (1-1.5 cm) of Black Baccara cultivar were cultured in VS media with various levels of BAP (0, 0.5,1, 1.5 and 2mg l-1) .shoot proliferation , highest stem and number of new leaves were measured. The maximum number of axillary shoots (3.56) and highest of stem (2.7 cm) was observed in the medium containing 1 mg/l BAP. Significant differences were not observed in the average number of new leaves between concentration of 0.5, 1 and 1.5 mg/l BAP. For rooting, three concentrations of VS mineral salts (full-, half, and quarter-strength) containing NAA (0.5 μM) were tested in semi-solid and liquid media. Tree strengths of VS (full. 1/2 VS, 1/4VS) were compared in sold and semi-sold mediums. The average number of roots (4.6) and root length (4.5 cm) observated in 1/4 VS. Generally, in all evaluated cases 1/4 VS medium was best than VS medium. Key words: Micropropagation; proliferation; in vitro culture; in vitro rooting; Rosa hybrida |
